Streptavidin/Avidin

Streptavidin is a tetrameric protein containing four biotin-binding sites, one on each subunit. The streptavidin/avidin-biotin system is one of the most widely used affinity-based technologies in biochemistry, molecular biology, diagnostics, and bioconjugation. The avidin/streptavidin-biotin interaction is among the strongest non-covalent interactions known in nature, with a dissociation constant (Kd) of approximately 10^-14 to 10^-15 M.

Because biotin can be readily introduced into proteins, antibodies, peptides, oligonucleotides, and other biomolecules, streptavidin serves as a versatile molecular connector that enables highly modular assay and conjugation platforms. Major applications of the streptavidin/avidin-biotin system include ELISA assays utilizing streptavidin-HRP detection, immunohistochemistry (IHC), nucleic acid detection, affinity purification, biomolecule immobilization on streptavidin-coated surfaces, and signal amplification technologies.
